Investigation of the Antiapoptotic Effects of Tofacitinib in C6 Glioma Cells Exposed to Oxidative Stress

This study aimed to investigate the potential antiapoptotic and cell-protective effects of tofacitinib in C6 glioma cells damaged by oxidative stress. The C6 glioma cell line was used in this study. Hydrogen peroxide was applied to the cells to induce oxidative stress. The cells were divided into 6 groups to receive control, H2O2, 100 nM tofacitinib, 50 nM tofacitinib, 25 nM tofacitinib, and 12.5 nM tofacitinib. The protective effect of tofacitinib application on cell viability and its effects on apoptotic processes were evaluated. Cell viability analyses were performed under a microscope, and caspase-3 expression was examined using immunofluorescence. Simultaneously, oxidative stress response was evaluated by measuring total oxidant status (TAS) and total oxidant status (TOS) levels.
Statistical analyses were performed using one-way ANOVA and Tukey post hoc test, with a p<0.05 value considered significant.
While H2O2 applied to the cells caused oxidative damage and apoptosis in the cell line, it was observed that cell viability was maintained and caspase-3 expression decreased in the groups treated with tofacitinib. Furthermore, a decrease in TOS levels and an increase in TAS levels were detected. These findings suggest that tofacitinib may reduce oxidative stress-related damage and have an anti-apoptotic effect.Tofacitinib was observed to suppress stress in c6 glioma cells subjected to hydrogen peroxide stress, preserving viability, and this is thought to be effective via an anti-apoptotic pathway. The findings indicate that tofacitinib may have neuroprotective effects in addition to its primary anti-inflammatory use.
