Abstract

Accurate and reliable evaluation of rumen fermentation is essential for improving ruminant nutrition, increasing feed utilization efficiency, understanding microbial processes, and controlling enteric methane production. For this purpose, different experimental methods are used in rumen research, which can be broadly classified as in vitro, in situ, and in vivo approaches. Among in vitro methods, batch gas production systems and the Rusitec system are the most widely used approaches, whereas in situ methods allow direct assessment of ruminal degradability, and in vivo methods enable evaluation of the overall biological response at the animal level. In this study, these three major methodological groups commonly used in rumen fermentation studies were comparatively evaluated. Batch gas production systems are rapid, cost-effective, and suitable for the preliminary screening of a large number of samples. The Rusitec system, on the other hand, offers important advantages for monitoring longer-term fermentation processes and microbial changes under more controlled conditions. In situ methods are particularly useful for determining ruminal disappearance and degradability characteristics of feeds, although they provide more limited information on fermentation end products. In vivo methods provide the highest level of biological relevance by linking rumen fermentation responses with animal performance, physiological reactions, and practical production conditions. However, they are also more demanding in terms of cost, labor, time, and experimental control. Overall, no single method appears to fully reflect the complexity of the rumen ecosystem on its own. Therefore, in vitro, in situ, and in vivo methods should be regarded not as alternatives that replace one another, but as complementary approaches serving different research objectives. In conclusion, selecting the most appropriate method according to the aim of the study, and combining these methods when necessary, may contribute to obtaining more reliable and interpretable results in rumen fermentation research.
